An algorithm based on the Braak histological staging procedure was applied to estimate Braak stages directly from the region of interest profiles in each subject. Quantitative region-based analysis of (18)F-AV-1451 images yielded region of interest and voxel level profiles that mirrored key features of neuropathological tau progression including profiles consistent with Braak stages 0 through VI.

Major finding: Older age predicted tau deposition in the medial temporal lobe and in the basal forebrain and insula, while patients with Alzheimer& Stage V includes pathology in additional cortical regions, including the superior temporal gyrus (STG, Brodmann Area 22). However, only NFT stage VI includes tau pathology in the primary visual cortex (VC, Brodmann Area 17, striate area).

Braak staging of tau

Braak staging, a framework for staging tau pathology in post-mortem tissue based on density and topology of tau, may now be used to stage tau pathology in vivo. The current proposal is designed to elucidate the temporal dynamics of AD pathology, especially tau, and associated functional connectivity changes in cognitively healthy older adults.

Taking the results a step further, Schöll worked out a method for in vivo Braak staging (see Braak and Braak, 1991). It defines seven stages of tau progression based on histopathology: no deposition in stage 0; the transentorhinal region in stage I; entorhinal cortex in stage II; fusiform and lingual gyri in stage III; neocortical association areas by IV. 2016-02-08 · The Braak staging scheme focused on neurofibrillary tangles, butabundant p-tau is also present in neuropil threads, and a recent scheme has been proposed by theBrainNet Europe consortium for staging tau pathology based on neuropil threads. 2016-03-02 · replicating the Braak staging approach to describe in vivo tau accumulation patterns. Our overall goal was to explore rela-tionships of the location of cortical tau ligand uptake with age, cognition, and Ab deposition—as measured by the Ab-binding ligand 11C-PiB (Price et al., 2005)—in cognitively healthy older Send correspondence and reprint requests to: Heiko Braak, MD, Clinical Neuroanatomy, Department of Neurology, Center for Clinical Research, University of Ulm, Helmholtzstrasse 8/1, 89081 Ulm, Germany; E-mail: heiko.braak@uni-ulm.de classification and Braak staging (Figure 2) (19). In these NIA-RI recommendations, Braak stages (transentorhinal, limbic and iso-cortical) are combined with the NP scores according to CERAD (infrequent, moderate and frequent) resulting in a statement of likelihood (low, intermediate, high) that dementia is due to AD-related lesions. The TauRx research team built on the findings of Braak and demonstrated a strong correlation between the degree of tau protein neurofibrillary pathology in the brain as measured using Braak staging, the amount of aggregated tau in pre-tangle brain regions and the degree of cognitive incapacitation as measured by scores on the mini mental state exam (MMSE).
